It is of vital importance to determine objectively the results of Wassermann reaction tests for syphilis. The colorimetric methods of determination that have been developed so far-one in which the color of the blood in the supernatant fluid of centrifuged Wassermann tests solutions is measured in order to estimate the degree of hemolysis taking place, or one developed by Kolmer and others in which the color of red-blood solution containing suspension of Wassermann tubes is compared with the colors of standard solutions for the purpose of estimating the capacity to impede the occurrence of hemolysis-involve complicated techniques and are not suited for practical applications. With a view to simplifying the objective method for reading the tests, the author has experimented on estimating the degree of extinction in electrophotometric determination of the results of Browning Wassermann tests for syphilis in 2,591 persons. The findings are summarized as follows: 1. There has been a correlation between the degree of extinction and the number of suspended red cells in Wassermann test solutions. It has been found that a regression line (or a curve) indicating the relation between them could be taken as a basis for determining the results of tests. 2. Strictly speaking, the degree of extinction in electrophotometric determination of Wassermann reactions is influenced by hemolysis factors. In practical application of this method, therefore, it would be advisable to use a red filter which eliminates the influence of hemolysis factors. 3. The concentration of sheep red cell used in Browning Wassermann tests is 3 %. However, too much importance should not be placed on the number of cells contained in it. The results of Ogata Wassermann tests (in which cardiolipin is used as antigen) could be determined more accurately for the number of red cells is comparatively small. 4. The degree of extinction in electrophotometric determination of the reactions occurring in solutions used for quantitative Ogata Wassermann tests (in which cardiolipin is used as antigen) has been equated with the quantitative titer obtained in Ogata quantitative Wassermann tests.
