This paper deals with the antimicrobial activities, food preservative effects and toxicity (chiefly chronic) of some naphthoquinone derivatives and methyl isopropyl phenol (Biosol). Among naphthoquinone derivaties, three compounds were used in this experiment, vitamine K_3, vitamine K_5 and M.N.Q(methylmercapto-1.4. naphthoquinone). The following results were obtained. A. Vitamine K_3. K_5 and M.N.Q 1. Acute toxicity: LD 50 in mice orally administrated was found to be 0.44 g/kg for vitamine K_3, 0.51 g/kg for vitamine K_5 and 0.89 g/kg for M.N.Q. 2. Chronic toxicity of V-K_3, V-K_5: Throughout 90 days experimental period, there was no appreciable difference between the control rats and those that received repeated oral administration of 1 mg/kg, 10 mg/kg, 100 mg/kg of V-K_3 and 10 mg/kg of V-K_5 as judged by the growth curve and final weight of several main intestinal organs. No pathological abnormalities were found in rats that received daily dosage of 1 mg/kg, 10 mg/kg of V-K_3 during the period of 90 days, but liver cell necrosis and fatty degeneration of the organs were observed in 100 mg/kg V-K_3 administration and even in 10 mg/kg V-K_5. 3. Chronic toxicity of M.N.Q : M.N.Q was found to have less toxicity than, that of V-K3. The animals received repeated administration in 100 mg/kg were tolerated. 4. Food preservative effects: It has been observed that V-K_3 was active against gram positive bacteria, some kinds of gram negative rod, fungi and yeasts at the concentration of 0.01-0.001 %. But it was common that such antimicrobial properties of V-K were inactivated by the presence of 1 % peptone. 5. Therefore this drug showed excellent effects in practical use to prevent spoilage Of protein foods, such as. Jam, Ketchup, Japanese bean jum, Shoyu and Tsukudani etc. 6. M.N.Q was demonstrated strong germstatic activity, and the activities was more effective, than that of V-K compounds. And the decrease of activities by peptone was less than that of V-K. B. Biosol. 7. Acute toxicity: LD50 in mice orally administrated was found to be 2.55 g/kg for biosol. No histopathological damage was observed at the concentration of 3.0 g/kg biosol in liver and kidney of experimental animal. 8. Chronic toxicity: In this case, the experiments were carried out throughout 180 days. The body weight increase of experimental animal were recognized normally as well as the control ones. And there were no pathological symptoms in gross appearance. It seemed that the less toxicity of biosol depended upon its insolubility. The histopathological examination of the organs are now in research.
