P-30 Isolation resveratrol from peanut root (Arachis hypogaea L.) and establishment of method for quantitative analysis

A phytoalexin, trans-resveratrol (3,4',5-trihydroxystilbene) was isolated from the ethylacetate soluble fraction from peanut root (Arachis hypogaea L.) by the repeated silica gel column chromatography. It was elucidated by the extensive spectroscopic analysis including MS, ^1H- and ^<13>C-NMR data. Optimum conditions for resveratrol quantitative analysis were determinded. The best conditon for solid phase, mobile phase and UV detection were Zorbax SB-C18 (4.6×150mm, 3.5μm), Water: MeCN (1%HOAc) and 254nm respectively. The equation of the regression coefficients for trans-resveratrol standard was y=0.001X+0.0021 (r^2=0.9994). Under these conditions, the value of RSD from peak area and retention time was 0.40%, 0.03%. The values of CV and accuracy in intra-day according to concentration of standard were 0.70%, 102.7%. trans-resveratrol content from peanut root was 1.7mg/g. These results might give useful information for recycling of peanut roots, since the isolated compounds have been known to have many beneficial activities such as antioxidative, anticancer and antimutagenic effects.
