44 新規骨格化合物blazeispirol類の単離と新規骨格形成機構に関する研究(口頭発表の部)

Blazeispirol A (Fig. 1. 1), the most abundantly produced secondary metabolite in the cultured mycelia of Agaricus blazei, is the first demonstration in support of the ring A-lost steroid skeleton in a living organism, and how is it synthesized only in the cultured mycelia is very interesting. In this paper, we report on the isolation and structure characterization of four new blazeispirane derivatives (2-5), four new protoblazeispirane derivatives (6-9) and two new ergostane derivatives (10, 11) from the cultured mycelia of Agaricus blazei. Compound 9 is seemed to be the actual intermediate of blazeispirol C. In order to characterize the expression gene which concern the biosynthesis of unprecedented skeleton, we tried the subtractive screening using the two different cultured mycelia as a RNA source. One of them, which is subcultured in flesh medium on three days interval, is not able to produce the blazeispirol derivatives. Another is the normal cultured mycelia which can produce blazeispirol derivatives (Fig. 4).
