In the course of screening program to discover new drug lead compounds from Brazilian medicinal plants, we investigated the constituents of bark of Brosimum acutifolium Huber (Moraceae), popular name "Murure". This plant has been used in folk medicine in the Amazonian region as an antiinflammatory and antirheumatic agent. The methanol extract of B. acutifolium was partitioned between ethyl acetate and water. The ethyl acetate extract was subjected to column chromatography on ODS and furthermore purified by HPLC on ODS. Seventeen new flavonoids (1-17) and one new lignoid (23) were isolated. Thirteen known compounds (18-22, 24-31) were also identified. The structures of these compounds were elucidated by spectroscopic methods. Fifteen (1-14, 18) of 22 flavonoids had isoprenoid side-chain at C-8. One (14) of the new flavonoids had a unique rearranged carbon skeleton and two (16, 17) of them were flavonolignans, each consisting of a stereoisomer of catechin and a phenylpropanoid. A new lignoid (23) had a novel allene structure. Thirty-one compounds from B. acutifolium were tested for inhibitory effects to protein kinase A (PKA) and C (PKC). Five compounds (8, 15, 17, 20, 29) inhibited PKA, 16 inhibited PKC, and 22 inhibited both PKA and PKC.
