The number, viability and oxygen consumption of alveolar macrophages were investigated in the free alveolar cells (FAC) harvested from rats by alveolar washing, with or without prior vascular perfusion, using several kinds of washing solutions differing in their content of divalent cations. Alveolar washing solutions free of Ca^<2+> and Mg^<2+> were most efficient in collecting the greatest number of FAC. When using solutions containing Ca^<2+> and Mg^<2+>, the cell yield decreased dramatically, indicating the role of these cations in promoting the adherence of the FAC in vivo. Prior vascular perfusion had a significant effect: when cations were absent from the perfusate solutions, the number of recovered cells inereased; when cations were present, the increase could still be obtained by the addition of EDTA on the alveolar side. The FAC viabiliy was above 90% except when solutions containing divalent cations were used. The oxygen consumption of cells obtained with physiological saline+EDTA was greater than with other solutions.
