Suppression of both 1st and 2nd meiotic divisions by caffeine treatment for the production of tetraploid Pacific abalone, Haliotis discus hannai (Archaeogastropoda : Haliotidae)

For the production of polyploid abalones by suppression of meiotic divisions, we adopted caffeine treatments in inseminated eggs from Pacific abalone, Haliotis discus hannai. Two procedures of a single treatment and two successive treatment with various exposure times were employed. In a single treatment of caffeine of 15mM, two experiments with two different exposed times gave similar results of producing polyploid cells with a triploid range of 54 chromosomes. In two successive treatments of caffeine, the 2nd treatments to eggs were performed at different times (4, 8, 12, and 16 min) after the 1st caffeine treatment. Each group of different treatment times allowed to produce polyploid cells with wide range in chromosome numbers from 15 to 101, with high frequency distribution of chromosome numbers of over 60. Among these polyploidies, cells with tetraploid (4n=72) and pentaploid (5n=90) were observed. These results suggested that the two successive treatment suppressed both the 1st and 2nd meiotic divisions of some eggs, while a single treatment suppressed only one of the divisions. Therefore, the two successive caffeine treatment is more effective than the single treatment. This study will provide some important information for future research on tetraploid induction in abalone.
