Four mammalian cultured cell lines, Indian muntjac IM, Chinese hamster CHL, HeLa and mouse L, were treated with a topoisomerase II inhibitor, etoposide at a concentration of 4μg/ml for 16 h in the presence of colchicine. Marked decrease in mitotic frequency was found in these cell lines with exception of CHL which showed an increased metaphase frequency compared to that of the untreated cells. A variety of chromosome aberrations including chromatid breaks, exchanges, abnormally separated sister chromatids and centric fusions were observed and the type of aberrations appeared characteristic to each of the cell lines used.
