The blood lactate was mainly produced from the working muscles which became hypoxia during exercise. The major evidence cited in support of general belief was that lactate concentration increased during submaximal exercise, and that this increase was less after training. Our purpose is to clarify on the level of ''biochemistry'' and ''molecular biology'' how exercise training can reduce the lactate production during workout. Experiments were performed on male rats, assigned to either untrained (n=6) and endurance trained (n=6) groups. Respiratory action of the isolated EDL and soleus was activated with addition of lactate much more in trained rats than in untrained ones. This evidence indicates that trained leg muscles can make lactate into a fuel through respiratory system. It is well known that LDH is a key enzyme in lactate metabolism, and that LDH-M and LDH-H catalyze reaction from pyruvate to lactate and from lactate to pyruvate, respectively. Total LDH activity was almost the same in EDL and soleus from trained and untrained rats, respectively. But LDH isozyme patterns were changed markedly by exercise training, increasing LDH-H subunit in both muscles on electrophoretic gel. These changes in isozymes were confirmed by amount of LDH B mRNA (corresponding to LDH M subunit) were much less in trained rat leg muscles than in untrained ones. Those results suggest the following fact:. The change in LDH isozyme pattern of muscle may reflect the adaptation in molecular level of metabolic process for effectine utilization of energy supply to muscle contraction.
