In order to elucidate the influence of high free fatty acidemia on the heart, intramyocardial gas tension (Pto_2, Ptco_2) was observed by using medical mass spectrometer (Medspect, MS-8, SRI) in male nongrel dogs before and after the intravenous infusion of FFA emulsion or physiological saline. Arterial blood gas tension (Pao_2, Paco_2 and pH) and hemodynamic parameters i.e. mean arterial blood pressure (MABP), aortic flow (AF), and coronary blood flow (CBF) were also observed. In addition, hearts were taken off 10 min. and 60 min. after the beginning of the infusion and studied by histochemical method. Results obtained were as follows ; (1) Pto_2 on both endocardial and epicardial sides were decreased remarkably after the infusion of FFA emulsion (non-incubated FFA), but Ptco_2 were not changed on both sides. On the other hand, neither Pto_2 nor Ptco_2 were changed after the infusion of FFA emulsion preincubated with bovine serum albumin (incubated FFA) or physiological saline. (2) MABP, AF and CBF were remarkably decreased after the infusion of non-incubated FFA, where as they were not changed after the infusion of incubated FFA or physiological saline at all. (3) As for the distribution of the Sudan III-stained substances in the heart, no difference was observed by the histochemical.analysis between hearts after the infusion of non-incubated FFA and those after the infusion of incubated FFA. Furthermore, bleeding, thrombi or emboli were not observed in both groups of hearts. From these results, it is concluded that high free fatty acidemia induces myocardial hypoxia and deleterious effect on hemodynamic parameters.
