It is well known that erythrocyte 2, 3-DPG content has a marked effect on the oxygen dissociation and is changing under various disease conditions. We studied on the effect of various steroid hormones on the levels of erythrocyte 2, 3-DPG. The results were obtained as following. 1) During pregnancy, 2, 3-DFG concentration gradually increased, especially in the third trimester was markedly elevated. While, during the menstrual cycle 2, 3-DPG concentration showed no significant change, but the highest concentration was seen in the luteal phase. In vitro, progesterone was found to increase the level of 2, 3-DPG after a 6 hour incubation. This effect was noted at 10^<-6> to 10^<-8> M of progesterone. Estrone, estradiol and estriol, however, had no effect. 2) Androgens (androsterone, dihydroteststerone, 5α-androstendiol and etiocolanolone) had no effect on 2, 3-DPG levels in vitro. 3) Hydrocortisone and prednisolone were found to increase the level of 2, 3-DPG, but dexamethasone and betamethasone had no effect. 4) Hydrocortisone direotly affected red cell metabolism and seemed to enhance the activity of diphosphoglycerate mutase. 5) In vitro study of blood which had been treated with hydrocortisone to elevate the 2, 3-DPG concentration revealed a shift to the right in the oxygen dissociation curve.
