The chromosomes of Eneoptera surinamensis were studied by C, NOR, and N (Mild and Strong) banding techniques. This species has 2♂=6+X_1X_2Y and 2♀=6+X_1X_1X_2X_2. The chromosomes of pair 1 have a proximal secondary constriction on their long arms, which is the only region marked by the silver staining technique in the whole complement. Mild N bands appear, barely visible, at the centromeric regions of pair 1 and at the region preceding the secondary constrictions of the long arms. Vestigial N bands also appear at the limit of the secondary constrictions, however, through the use of the Strong N banding technique these vestigial bands appear well defined. The colloidal Ag and N banding techniques did not mark the same chromosomal regions, but they appeared very close to each other, indicating that the N bands are composed of non-histonic proteins associated to the NORs. All the chromosomes of the complement, with the exception of the X_2, have pericentromeric C bands. The chromosomes of pair 1 have two additional markings on the interstitial regions of their long arms, while the Y shows two interstitial blocks near the pericentromeric C band and another two near the telomeric regions. The pericentromeric C bands of the X_2 chromosomes might have been lost after the fusion with its ex-homologues, actually the Y chromosome, since it has similar dimensions to one of the arms.
