Key roles of condensins in mitotic chromosome assembly and segregation(The Society of Chromosome Research Award Lecture)

The faithful segregation of duplicated genetic information into two daughter cells is central to cell proliferation in all organisms. To this end, chromatin undergoes progressive structural changes during the cell cycle. At the onset of mitosis, duplicated DNA molecules are converted into "metaphase chromosomes" composed of a pair of cylindrical structures called sister chromatids. During the past decade, a number of approaches have been combined to demonstrate that a protein complex known as condensin (henceforth condensin I) is a key player in mitotic chromosome assembly. More recently, we identified a second condensin complex (hereafter condensin II) from vertebrate cells. While condensins I and II display different kinetics in chromosomal association, they cooperatively contribute to global and regionspecific architecture of metaphase chromosomes, and ensure proper chromosome alignment at the metaphase plate, followed by sister chromatid separation in anaphase. Apart from their role in mitosis, the condensin complexes may have important roles in interphase processes such as genome organization and stability, implicating their fundamental and broader roles in chromosome metabolism. Thus, chromosomes could have additional roles beyond the transportation of DNA. Furthermore, the discovery of condensin II provides evolutional implications in chromosome structure. Finally, I also summarize recent progress in our understanding of molecular basis of human hereditary diseases associated with defects of the mitotic chromosome assembly.
