To determine age-related decline in memory function in the rat by the longitudinal method, a group of male Wistar albino rats were tested on a spatial delayed nonmatching-to sample task at the ages of 5 months (youth) and 30 months (senescence). A test trial consisted of sample run and choice run. In the sample run, the rats were forced to enter a pre-determined arm of a two-arm radial maze and, in the subsequent choice run, were allowed to choose between the two arms. The selection of the arm not entered during the sample run (nonmatching-to-sample) was rewarded as a correct response. After having attained high accuracy of performance in the task with 5 sec delay interposed between the sample and choice runs, the animals were tested with prolonged delays of 20,80,320 seconds. The animals, at both ages, showed the perfect accuracy (100% correct) for 5 sec delay but delay-dependent decline in accuracy for the prolonged delays. The rate of delay-dependent decline at the age of senescence was not different from that at the age of youth. The negative finding from the present study was compared with the positive findings from previous cross-sectional studies and some factors responsible for the discrepancy were discussed.
