Nucleolar organizer regions(NORs) of normal rat bone marrow cells, fibroblasts, and 9 erythroblastic leukemia cell lines were studied by Ag-NOR staining and in situ hybridization methods, Normal bone marrow cells in vivo and fibroblasts in vitro had an average of 5.6 and 5.8 Ag-positive NORs (Ag (+) NORs) for 6 NORs per metaphase, respectively. Growth stimuli given 24 h before harvesting of bone marrow cells slightly increased the incidence of Ag (+) NORs. Ag (+) NORs in normal bone marrow cells in vitro decreased in number as time passed. Leukemia cells had a lower number of Ag (+) NORs than the normal bone marrow cells and fibroblasts. The distribution of Ag (+) NORs among NORs in leukemia cells was abnormal and different from case to case. Furthermore, the distribution of Ag (+) NORs was different from that of ribosomal cistrons shown by the in situ hybridization method. Interphase nucleoli were much more heavily labeled with AgNOR grains. The abnormal increase and distribution pattern of ribosomal DNA and abnormal pattern of their activity in leukemia cells were discussed.
