A perfusion system of isolated rat femur was established to study the response to PTH by adenosine 3', 5'-monophosphate (cAMP) release. Synthetic bovine PTH 1-34 was significantly more potent than bovine PTH 1-84 to stimulate cAMP release from the perfused bone on the equimolar basis. Bones form younger rats (6-week old) were more sensitive to PTH 1-34 in the cAMP response than older (16-week old) ones. Females were more responsive to PTH 1-34 than males. These results suggest that isolated rat bone perfusion system is a useful experimental model for the study of PTH action on the bone.
