In order to assess the interaction between parathyroid hormone (PTH) and vitamin D_3 metabolites on the hormone receptor renal adenylate cyclase system, renal tissue samples obtained from wister rats were examined for the c-AMP content. Renal c-AMP content was measured by the competitive protein binding assay, and compared before and after the intravenous administrations of PTH, 1α(OH)D_3 1,25(OH)_2D_3 and these various combinations in normal and vitamin D deficient female wister rats. The results were as follows : 1) The c-AMP content of normal renal tissue increased 1.85 times one minutes after the administration of PTH. 2) The c-AMP content of normal renal tissue showed no increase after the loading of 1α(OH)D_3 or 1.25(OH)_2D_3. It responded to PTH 60 minutes after the 1α(OH)D_3 pretreatment, and its increasing ratio 1.92 was almost as same as that of PTH alone administrated. 3) The renal c-AMP content of vitamin D deficient rat showed no increase 1 minutes after the administration of PTH alone. (Increasing ratio was 1.10), but it showed some response 1 minutes after 1,25(OH)_2D_3 was given. (Increasing ratio was 1.43). 4) The renal c-AMP content of vitamin D deficient rat showed as same increase to PTH administration 60 minutes after the pretreatment of 1α(OH)D_3, and 5 minutes of 1,25(OH)_2D_3 as noted with PTH alone in normal kidney. (Increasing ratio was 1.74 and 2.12). These findings suggest that vitamin D does not enhance the reactivity of renal adenylate cyclase to PTH in normal rat, but in the vitamin D deficiency, renal adenylate cyclase little respond to PTH, though its reactivity come to restore after the vitamin D deficiency was corrected by administrating vitamin D_3 metabolites.
