Serological ABO blood group typing is a general method for determining blood types as AB, A, B or O. This paper, presents a method for convenient genotyping to determine ABO blood type using polymerase chain reaction (PCR) and single-strand conformational polymorphism (SSCP) in conjunction. PCR amplified products from exon 6 of the ABO gene were each found to move differently on polyacrylamide gel in SSCP. Genes for A, B and O alleles of O1 and O2 were clearly separated on the gel. Allotypic allele combinations representating A/B, A/O1, A/O2, B/O1, B/O2, O1/O1 and O1/O2 could be genotyped with a single panel. This simple PCR-SSCP method may be used to distinguish O1 and O2, thus making available more information on ABO blood group genotyping. This method should prove useful in the laboratory.
