Effect of OPC-21268 on Cytosolic Free Calcium in Cultured Rat Mesangial Cells

The effect of nonpeptidic V_1 receptor antagonist OPC-21268 on cellular action of arginine vasopressin (AVP) was examined in cultured rat mesangial cells by measurement of cytosolic free calcium [Ca^<2+>]i) and V_1 receptor binding study. OPC-21268 inhibited AVP-induced increase in [Ca^<2+>]i in a concentration-dependent manner. In contrast, an angiotensin II-induced increase in [Ca^<2+>]i was not affected by pretreatment of the cells with OPC-21268. OPC-21268 caused a concentration-dependent displacement of [^3H] AVP binding to V_1 receptors, and Lineweaver-Burk analysis revealed that OPC-21268 competitively antagonized V_1 receptors. These results suggest that OPC-21268 acts as a specific V_1 receptor antagonist against the contractile action of AVP in mesangial cells.
