国立情報学研究所で電子化To elucidate the functions of pancreatic endocrine glands, we investigated the pharmacokinetics of full-sequence and truncated form of glucagon-like peptide 1 (GLP-1) in vivo, using radioimmunoassay for each peptide in normal and storeptozotocin (STZ) diabetic rats. The half-disappearance times (T1/2) and metabolic clearance rates (MCR) of truncated GLP-1,given as a bolus injection, were, respectively, as follows : T1/2 (min) 2.7±O.7 (mean±SD) and 5.9±1.5 ; MCR (ml・kg^-1・min^-1) 51.9±21.5 and 66.O±25.5. The T1/2 and MCR values were, respectively, shorter and larger than those of full-sequence GLP-1. The T1/2 and MCR values of constant infusion of truncated GLP-1 in STZ diabetic rats were, respectively, 3.1±1.1 min and 68.3±22.6 ml・kg^-1・min^-1,and these values were not different significantly from the respective values found in normal rat. These findings demonstrate that truncated GLP-1 degrades faster than the full-sequence form and may be an active form of GLP-1 in vivo.
