マウス皮下腫瘍モデルにおけるT細胞選択的サイトカイン,IL-2 Selective Agonist,を用いた抗腫瘍効果の基礎的検討

Interleukin(lL)-2は進行した転移性癌に対して有効な抗腫瘍効果を持つとされている。ただしその使用は,容量依存的に引き起こされるvascular leak svndrome(VLS)などの副作用により制限されることが多い。IL-2による副作用の原因の一つとしてNK細胞の直接的な活性化があると考えられている。このような副作用を抑えるためにrhIL-2のアミノ酸を一ヵ所改変し,主にNK細胞に発現している中親和性のIL-2レセプターとの結合を起こりにくくしたIL-2 selective agonist(IL-2SA;BAY50-4798)が開発された。そこで,われわれは今回,IL-2SAがrhIL-2の全身投与と同等の抗腫瘍効果を持つか否かをマウス腫瘍モデルを用いて比較検討した。マウス皮下腫瘍モデルにおいて,IL-2SAの全身投与は,rhIL-2と同様の腫瘍増殖抑制効果を示すと共に,高い細胞障害性Tリンパ球活性を示した。さらに,IL-2SAはrhIL-2に比較しNK細胞を活性化する作用が弱かった。また肺における血管透過性の亢進を抑えることができた。以上より,IL-2SAは抗腫瘍効果はIL-2と同等と考えられ,さらにIL-2に比較して副作用の弱いことが確認できた。したがってIL-2SAの全身投与は進行癌に対する有効な治療法として期待できる。
