The purpose of this experiment was to find an index for intraoperative ischemic-reperfusion injury in the cirrhotic liver. The rats were divided into N group with 60 minute ischemia in normal livers, and CL group with 60 minute ischemia in cirrhotic livers. 1%Dimethylnitrosamine was administered intraperitoneally to CL group of Wister rats on three consecutive days of each week for a period of four weeks. Digital videotapes recorded with a pencil lens-probe charge-coupled device microscope were analyzed with NIH Image software. In zone 3, the sinusoid diameter (DS=sinusoid diameter of zone 3) was measured and the volume fraction (Vv=volume fraction of zone 3) were calculated in preischemia and after 10, 20, 30, and 60 minutes of reperfusion. The Ds was significantly shorter in CL group than in N group at each point. Vv in CL group at 10 minutes after ischemia was significantly less than that preischemia but not in N group. Vv were significantly less in CL group than those in N group at each point from preischemia to 60 minutes of reperfusion. Survival rate was 0 % in CL group and 100% in N group. In conclusion rats with DMN-CL didn't recover well during reperfusion after 60 minutes ischemia in vivo microcirculation and didn't survive.
