Interleukin-1α Production by Cultured Human Decidual Cells

Objective : To investigate the effect of various inflammatory cytokines on interleuhin(IL)-1α synthesis in human decidual cell cultures.Methods : Decidual cell cultures were stimulated with various concentrations of tumor necrosis factor(TNF)α, IL-1β, and lipopolysaccharide(LPS).At the same time, a protein kinase C(PKC)activator 12-0-tetradecanolylphorbol 13-acetate(TPA), or a PKC inhibitor 1(5-isoquinoline-sulfonyl)-2-methylpiperazine dihydrochloride(H-7)was added to the cultures.IL-1α concentrations in the cell lysate and prostaglandin(PG)E_2 concentrations in the supernatants were measured.Results : (1)When human decidual cell cultures were stimulated with TNFα, IL-1α synthesis increased in a dose-and time-dependent manner.(2)IL-1α synthesis was also enhanced by IL-1β or LPS stimulation.(3)When the cells were co-treated with TPA and TNFα, the TNF-α-induced IL-1α synthesis was inhibited.(4)Treatment with TPA alone did not inhibit IL-1α synthesis.(5)When the cells were co-treated with TPA and IL-1β or LPS, the IL-1β- or LPS-induced IL-1α synthesis was also inhibited.(6)H-7 treatment did not inhibit TNF α-induced IL-1α synthesis.(7)H-7 treatment did not abolish the inhibition of IL-1α synthesis induced by co-treatment with TPA and TNFα.(8)Co-treatment with TNFα and TPA produced a greater increase in the concentration of PGE_2 than did single treatment with TNFα or TPA.(9)H-7 treatment inhibited the enhanced PGE_2 secretion induced by TPA.Conclusion : The present results suggest that TNFα-stimulated IL-1α synthesis in human decidual cells is inhibited by TPA, a PKC activator, that may be due to a PKC-independent pathway.Enhanced IL-1α and PGE_2 secretion induced by TNFα may cause contraction of uterine muscle and is speculated to be a risk factor that provokes spontaneous abortion.
