The experimental periodontitis of mice infected by Porphyromonas gingivalis (Pg) has been developed in our laboratory. The aim of this research is to elucidate immunoresponses in the sera and periodontal tissues after the experimental periodontitis is induced with Pg. For this purpose, the serum antibody titer and the number of T- and B- cells in the Pg-infected gingival tissues were measured. Male C3HIHeN mice at 7 weeks old were divided into either an infected or control group (a non-treatment group at zero week was also included in the control group). The serum antibody titers were measured by the ELISA method. The CD3 and CD20 cells in the periodontal tissues were examined with the immunohistochemical method. For the infected mice, the serum IgM titers against the ceH walls of Pg were significantly higher than those of the control group at 3 weeks after the treatment. On the other hand, the serum IgG titers in the infected group significantly increased at 5 and 7 weeks. Serum IgA titers in the infected group significantly increased at 7 weeks. The number of CD3 positive cells in the epithelium of the control group was significantly larger than that of the infected group at I week. Moreover, the number of CD3 positive cells in the connective tissue of the infected group was larger than that in the epithelium at 5 and 7 weeks after the treatment. The number of CD20 positive cells in the connective tissue of both groups was larger than that in the epithehum at 5 and 7 weeks. This model may be useful for analyzing the immunological res ponses in the sera and periodontal tissues.
