The purpose of this study was to examine destructive and reparative changes of the periodontal tissue especially alveolar bone during mechanical imapaction and the removal of impacted material. Eight-week-old S.D. rats were devided into the compression group and the removed group of compression. In the compression group, a guttapercha point was inserted into the interdental papilla between the upper two molars (M_1 and M_2). In these animals, destructive changes of the periodontal tissue were histopathologically observed at intervals of 1,3,5,7 days (1 week), or 14 days (2 weeks) after the insertion. In the group in which compression was removed, the guttapercha point was removed after 1,3,5,7 days (1 week) or 14 days (2 weeks) of insertion, and reparative changes of the periodontal tissue, in particular the alveolar bone, was histopathologcally observed at intervals of 1,3,5 days, 7 days (1 week), 14 days (2 weeks), 28 days (4 weeks), 56 days (8 weeks) or 84 days (12 weeks) after the removal of inserted material. In addition to the microscopic method in the removed group of compression, the specimens were observed by the fluorescence labeling method. Compression group : One day after compression, the interdental pailla was destroyed and depressed. As a result, disappearance of the epithelial layer, inflammatory cell infiltration, and disappearance of transseptal fibers were observed in the papilla. Three days after compression, inflammatory cell infiltration was severe, transseptal fibers had almost disappeared, and bone resorption due to osteoclasts from the lateral side of the septum was observed. One week after compression, inflammatory cell infiltration was slight, and regeneration of transseptal fibers and epithelium layer was observed. Two weeks after compression, interdental papilla consisting of transseptal fibers was clear, bone resorption was in progress in the interalveolar septum. Compression removed group : In animals in which compression was removed one day after the beginning of compression, interdental papilla was restored to its original from after one week. The findings of the specimen removed after compression for three days resembled those of the specimen removed after compression for one day, and regeneration of transseptal fibers after five days. Repair of interalveolar septum by the osteoblast was observed after two weeks, but the epithelial portion was located on the lower root surface from the enamel-cement junction. In the group in which compression was removed after five days, the interdental papilla was restored to its original form with the repair of transseptal fibers after one week, and repair of the alveolar bone was insufficient. Although the height of the interalveolar septum remained reduced, bone was observed on the lateral side after two weeks. In the group in which compression was removed after one week, the interdental papilla was restored to its original form after three days, the interalveolar bone was recovered with the width after two weeks, but the epithelial attachment portion was located on the lower root surface from the enamel-cement junction. In the group in which compression was removed after two weeks, the course was similar to that in which compression was removed after one week was observed.
