ECTOPIC EXPRESSION OF LUMICAN IN LENS EPITHELIUM DURING WOUND REPAIR

Background and Purpose: Lumican is a member of the keratan sulfate proteoglycan (KSPG) protein of small leucine-rich proteoglycan family (SLRP), which is also ectopically and transiently expressed by migrating and restratified epithelial cells following corneal epithelium debridement. It was suggested that the expression of lumican mRNA and protein may facilitate corneal epithelial wound healing (Saika S, et al.,JBC 275: 2607-2612,2000). In the present study, we examined the expression of lumican in healing, injured, lens epithelium of mice and humans. Methods: The crystalline lens of C56B16 mice (n=42) was punctured and allowed to heal up to 3 months. Human lens epithelia freshly obtained during cataract surgery (n=3) and also those (n=21) secondarily removed after interval up to 10 years after the initial operation were included in the study. The specimens were processed for immunohistochemistry for lumican and KSPG (5D4). Results: The expression of lumican was up-regulated 24 hr after a puncture injury. At later stages, extracellular matrix accumulation was detected beneath the capsule in association with lens cell proliferation. It was found to be labeled by anti-lumican antibodies. Matrix accumulation in human specimens was also found to be positive for both lumican and KSPG, whereas the fresh lens epithelium was negative for lumican. Conclusion: Human and mouse lens epithelia up-regulate the expression of lumican during healing. Lumican may serve as a marker protein of healing, injured, epithelial tissue.
