ALTERED EXPRESSION OF THE LYSYL OXIDASE-LIKE (LOXL) GENE COINCIDES WITH ABNORMAL HAIR FOLLICLE DEVELOPMENT IN ROUGH COAT (RC) MICE

Lysyl oxidase-like protein shares close homology within its C-terminal end with the active form of lysyl oxidase and has been shown to have in vitro catalytic activity towards collagen and elastin substrates. Our previous studies demonstrated co-expression of the LOXL and collagen typeIII genes and detected LOXL associated with collagen fibers in the dermis. We have mapped the mouse LOXL gene to chromosome 9. In subsequent back crosses using an intragenic RFLP with BALB/C and C57BL/6 strains, the LOXL gene co-segregated with markers D9Mit232 and D9Mit211 located at 31 cM on the standard linkage map of mouse chromosome 9 close to the RC locus. The RC phenotype abnormalities include small body size, cardiac congestion and intensive hair loss resulting from multifocal hair follicular atrophy and abnormal hair shaft formation. We tested the potential involvement of mutations of the LOXL gene in the RC phenotype. The LOXL mRNA was detected in most tissues but at significantly reduced level in the heart of RC mice. Immunostaining of LOXL in normal mouse skin sections detected intensive staining in the epidermis, dermal papillae, follicular root sheaths, and sebaceous glands. The connective tissue in the dermis, basement membrane of the adipose tissue and muscle layers were also stained. In the skin of RC mouse LOXL immunostaining was reduced in intensity in deformed and atrophic hair follicles and hypertrophic sebaceous glands. The skin sections in LOXL knockout mouse detected no staining but exhibited normal morphology of the hair follicles. These findings indicate that reduced LOXL expression may not be directly but indirectly associated with mutations related to abnormal hair follicular development of the RC gene.
