To elucidate the morphogenesis, epithelium differentiation, myogenesis, and vasculogenesis of the tongue, Sprague-Dawley rat embryos were examined with scanning and transmission electron microscopes. Some of these specimens also were prepared for observation with the light microscope. The first sign of tongue development was a slight swelling on the inner end of the first branchial arch at embryonic day (e.d.) 13. At e.d.14,the primordium increased in size and already possessed fundamental tongue morphology. Microvilli usually were found on the free surface of the superficial epithelium of the lingual dorsum before e.d.19. These microvilli might act as a reservoir of cell membrane required for rapid tongue growth. On the other hand, at e.d.19.5,well-developed microridges were observed on the free surface of the lingual dorsal epithelium, the cells of which formed a continuous epithelial sheeth and covered the dorsal lingual surface. At e.d.19.5,an unusual epithelial cell layer without keratinization was found. This layer coverd the surface of the tips of the well-developed keratinized filiform papillae, and was lost from the lingual dorsal surface at e.d.20. Aggregation of the mesenchymal cells was observed in the tongue at e.d.15.5. Some of these mesenchymal cells possessed a few myofibrils, suggesting the initiation of myogenesis. At e.d.18,in each lingual muscle, myofibrils of the myotubes increased in number and their nuclei began to move to the periphery of the cells. Each lingual muscle had a well-developed mature appearance by e.d.19. The blood vascular system was immature until e.d.18. The wall of the blood vessels consisted of only endothelial cells. At e.d.19,mesenchymal cells closely related to the periphery of the endothelial cells often were observed. These findings suggest that immature blood vessels differentiate into true capillaries with pericytes, or into arterioles or venules after e.d.19.
