Periodontitis is a chronic inflammatory disease associated with connective tissue destruction caused by an infection of Gram negative anaerobes. It is well known that T cells are functionally activated to regulate immune responses in periodontal lesions ; however, the immune recognition of antigens of periodontally-associated bacteria by these T cells has not been elucidate. The aim of this study is to characterize the immune recognition of Pg antigen fractions by naive T cells. Antigens from Porphyromonas gingivalis FDC 381 whole bacterial cells (PgWC) were solbilized in SDS-PAGE sample buffer and were fractionated in a molecular weight range of 14.0-30.0 kDa by using SDS-PAGE. Proliferation induction activity (PIA) of each fraction was assessed as a relative amount of BrdU incorporated into peripheral blood mononuclear cells derived from 6 healthy subjects in stimulation culture with the fraction. One of the subjects who showed higher PIA against Pg possessed higher PIAs against fractions containing 20.5,26.5 and 28.5-30.0 kDa proteins, another subject had all of the fractions (14.0-30.0 kDa) tested. However, one of the subjects who showed lower PIA against Pg possessed higher PIA against fractions containing 18.0-20.0 and 28.5-30.0 kDa proteins, and another did not possess high PIA against any of the fractions. In the early phase of stimulation culture (3 days), significant positive correlation (p<0.01) was observed between PIA of 30.0 kDa fractionsand that of PgWC. In 5 days of stimulation culture, positive correlations (p<0.05) were observed between PIA of fractions in 18.0-20.5,27.0,28.5 or 29.0 kDa and that of PgWC. Positive correlation (p<0.05) was observed between PIA of fractions in 20.0-20.5,28.5 or 29.0 kDa in 5 days of stimulation culture and that of PgWC in 7 days of culture. It is suggested that PgWC-reactive T cells consist of several clones which recognize various Pg antigens. The individual differences in the antigen recognition of those T cell clones might play a critical role in individual pathogenesis of periodontal disease.
