When the exponential growth phase cells of Candida albicans were cultured in GN medium containing thiazolidine-4-carboxylic acid (thioproline), proline, Sodium bicarbonate, Sodium acetate, Sodium phosphate and N-acetyl-D-glucosamine at 37℃ for 3h, over 70% of the cells formed germ tubes (GT). In these experiments, the substances in GN medium were examined for induction of GT formation. When the starved exponential phase cells were precultured in the thioproline containing medium, then were cultured in the zinc medium which thioproline in GN medium was replaced by ZbSO_4,over 70% of the starved cells formed GT. However, under 15% of the starved cells precultured in the medium without thioproline formed GT in the zinc medium. The results in this study showed that only thioproline could induce GT formation, while N-acetyl-D-glucosamine could not. Cysteine and methionine were also effective for induction of GT formation, when thioproline in GN medium was replaced by cysteine or methionine.
