抗癌剤が可移植性ヒト口腔癌切除後の残存腫瘍に及ぼす影響

The histopathology, nuclear DNA contents and changes of a cell cycle of transplantable human oral cancer, removed partially, with and without cisplatin (CDDP) or peplomycin (PEP) administered alone or in combination with both drugs were investigated with flow cytometry (FCM) and H. E staining for 15 days after transplantation. The following results were obtained; 1. In control tumors excised partially, the relative tumor weights and S phase cell populations significantly increased at 10,12 and 15 days, and at all days whereas, G_0/G_1 phase cells were significantly reduced at all days. 2. Body weight of tumor-bearing nude mice in each group administered i. p. CDDP (4 mg/kg) or PEP (17 mg/kg) alone once a day for 5 days was significantly reduced at 3,5 and 8 days after treatment. In the group administered in combination with both CDDP (4 mg/ kg) with one shot and PEP (8.5 mg/kg) once a day for 5 days, body weignt was significantly reduced from 1 day after PEP administration. 3. In each group administered i. p. CDDP or PEP with one shot, the relative tumor weights were significantly reduced in a group at a dose of 4 mg/kg of CDDP from 1 day and for the other groups from 3 days, whereas it was significantly reduced in the group administered i. p. CDDP or PEP alone once a day for 5 days from 3 days. They became the smallest in the group at a dose of 4 mg/kg of CDDP at 8 days after treatment. In the combination group with two drugs, were significantly reduced from 1 day. 4. In the groups administered CDDP at doses of 2 and 4 mg/kg with one shot, S phase cell populations significantly increased from 1 day at both doses and G_2+M phase cells from 3 days at a dose of 4 mg/kg. In each group administered CDDP at doses of 2 and 4 mg/kg once a day for 5 days, G_2+M phase cells significantly increased from 3 and 1 days, whereas G_0/ G_1 phase cells were significantly reduced from 1 day. 5. In the groups administered PEP (8.5 mg/kg) with one shot and once a day for 5 days, G_2+M phase cell populations significantly increased at l and 2,but not other, days. In the group administered PEP (17 mg/kg) with one shot and once a day for 5 days, G_2+M phase cells significantly increased from 1,5 and 8 days, whereas S phase cells from 3 days. 6. In the combination group S phase significantly increased from 3 days and G_2+M phase cells at 1,3 and 5 days after treatment. 7. Grade III was histopathologically observed in the groups administered i. p. CDDP (4 mg/kg) alone and in combination with both drugs, grade IIb in the group given i. p. PEP (17 mg/kg) and grade IIa in the groups i. P. given i. p. CDDP (2 mg/kg) and PEP (8.5 mg/ kg) at 5 days after treatment.
