歯牙欠損がラットの細胞性免疫機構に及ぼす影響 : 胸腺, 脾臓および顎下リンパ節の生細胞について

This study was designed to determine the effects of teeth defects on the cell-mediated immunology of rats. The rats were divided into four (A〜D) groups; A groups without any treatment, B groups pair-fed with experimentals, C groups subjected to extraction of lower right three molars and to lower incisal teeth amputation twice every three days, and D groups subjected to the same protocol with C groups and injected with a β_1-adrenergic blocker, metoprolol (l0mg/kg) once a day for 7 days consecutively. Two T cell subsets (W3/25 and OX8) and IL-2 receptor in thymus, spleen and submandibular lymph node freshly obtained from four groups were analyzed by the ABC method histologically and flow cytometrically at 7 days after various treatment. The following results were obtained : 1. Body weight of B and two experimentals (C and D groups) was significantly decreased but that of the D group recovered up to that of the B groups. 2. Wet weights of thymus tended to reduce in contrast that those/100g body weight did to increase in the B groups and two experimentals. 3. Wet weights of spleen in the B and C groups were significantly reduced in contrast that wet wights and those/100g body weight in the D group were significantly increased. 4. The three antigen (W3/25,OX8 and IL-2R)-positive T cells were observed in the three tissues of each group by the ABC method. 5. In thymus the double-positive (W3/25^+ OX8^+, D. P.) T cell subsets were significantly reduced in contrast that T helper (W3/25^+ OX8^- , Th) cells were significantly increased in the B group and two experimentals. In addition, T suppresser (W3/25^- OX8^+, Ts) and double-negative (W3/25^- OX8^-, D. N.) T cells were significantly increased in the B and C groups. 6. In spleen, Ts cells were significantly reduced in contrast that D. N. T cells were sugnificantly increased in two experimentals. 7. In submandibular lymph node, D. N. cells were significantly increased in the C group in contrast that in the D group these cells were significantly reduced. In addition, Th cells were significantly increased in the B and D groups. 8. The IL-2R positive cells were saignificantly reduced in W3/25^+ T cells of thymus and in W3/25^- T cells of spleen in two experimentals. 9. The ratio of W3/25^+ to OX8^+ in thymus analyzed by single color analysis was significantly increased in the B group and two experimentals. In addition, the ratio of Th/Ts in thymus was significantly increased in the D group.
