Dynamic Properties of Protein and Water Studied Through Effects of Water Content on Water Relaxations in Erythrocytes

Magnetizations of water protons in human erythrocytes were measured using an off-resonance irradiation technique by varying the irradiation frequency and water content. The frequency dependences of the magnetizations were well expressed by the rate equations, which gave the tumbling time of hemoglobin protons and the intrinsicrelaxation rates of water and hemoglobin protons. Analysis of the data determined the amounts and the average correlation times of bound water fractions. The average lifetime of the bound water in the interior of the protein with respect to exchange with the bulk water was found to be 1.0 ± 0.3 μs. The estimation of the surface hydration layer showed excellent agreement with the amount measured by thermodynamical methods.
