Blasticidin S, an antibiotic effective against rice blast disease, is irritative to skin, mucous membrane and also induces severe diarrhoea when inhaled or it gets into human body from the oral cavity. It is, therefore, important to analyse its effects to human body, from the view point of health supervision of blasticidin S workers or users. This study deals with the relation between the distribution and the velocity of excretion of blasticidin S in the organs and in the blood of mice by the tracer technique after oral and intravenous administrations and histopathological examination, with discussion about the result of the health examination of blasticidin S workers. Summary is as follows. (1) Screening tests on blood and urine were carried out on workers. The presnt results showed no abnormal findings which could be clearly attributed to the toxicity of blasticidin S. (2) Tritiation of blasticidin S was done by Wilzbach's method. Purification of tritiated blasticidin S was carefully done and indentified by the physiochemical and biological methods. The product has a specific activity of 106μci/mMol. (3) The radioactivity of tritium in the blood and organs was measured after oral and intravenous administrations of tritiated blasticidin S to the mice. (4) Most of blasticidin S was rapidly excreted within 24 hours. (5) The accumulation of blasticidin S in any specific organs was not observed in the case of the oral administration, but the high concentration of it in the kidney was seen in the case of the intravenous administration. (6) In observations of the microautoradiography technique, the tritium grains were seen in the uriniferous tubules in the kidney, some of the white pulps in the spleen, the villi, the lamina propria mucosae and the submucosa in the intestines, and the lobules in the liver. (7) Histopathological observation was as follows. The effect of blasticidin S by subcutaneous injection seems to be manifested in the proximal convoluted tube of the uriniferous tubules and in the skin. No effect was observed in other organs. (8) The oral administration to mice over a long period at a low concentration showed no marked or characteristic changes in various organs except for edema of the lamina propria mucosae and the submucosa in the intestines. (9) Acute toxicity of blasticidin S at a high concentration was tested. Histopathological examination showed marked edema of the lamina propria mucosae and the submucosa in the intestines. (10) It is concluded that the unfavourable effect of blasticidin S on the human body used to the agricultural drug is only severe contact inflammation of the mucous membrane or skin and neither acute nor chronic harmful changes occur on the parenchymatous organs.
