Pretreatment of Coleus blumei (BENTH.) stem explants for 24 hr with L-proline (P) (1.0mM) by the successive cup technique resulted in the redifferentiation of extremely high numbers of wound vessel members (WVM) from pith and cortical parenchyma tissues following the addition of a xylogenic concentration of 5 ppm indoleacetic acid (IAA). The addition of P with auxin or after auxin had been introduced into the segments was ineffective in altering the numbers of WVM formed as compared to controls. P-treated segments showed both normal and aberrant WVM with the addition of colchicine (0.01%) and IAA; colchicine-treated segments invariably produced aberrant WVM when P and IAA were given. These results suggest that a P-rich substance may interact with auxin in initiating wound vessel member differentiation.
