Swelling of spinach chloroplasts induced by HCHO and KMnO_4, an effect partially hidden by the wide size range of chloroplasts

Both KMnO_4 and HCHO in concentrations used for fixation for electron microscopy induce pronounced swelling of spinach chloroplasts. However, since electron microscopy samples small numbers, it is possible to overlook the swelling effect because the size range of the swollen chloroplasts can overlap the extremely wide range of chloroplasts in living mesophyll cells. HCHO fixes spinach chloroplasts only after 16 hr incubation, as verified by failure of the chloroplasts to swell when subsequently washed with water. However, the HCHO treatment fails to prevent an initial swelling and KMnO_4 further swells chloroplasts pre-fixed with HCHO. Spinach chloroplasts in vivo measured in face area 27.7±0.06μ^2 mean value, 23.8μ^2 mode value, range 6.2 to 10_2.9μ^2, and their distribution is skewed so that the coefficient of skewness is 0.15. Chloroplasts isolated directly into phosphate buffered 4% HCHO after 24 hrs measured in face area 58.2μ^2 mean value, 46.5μ^2 mode value, range 22 to 121μ^2, and the coefficient of skewness increased to 0.24. When such chloroplasts were additionally treated with phosphate buffered 2.8% KMnO_4 the spinach chloroplasts measured in face area 96.4±1.40μ^2 mean value, 86.1μ^2 mode value, range 22 to 203μ^2, and the coefficient of skewness unchanged at 0.24. Volumes of spinach chloroplasts isolated in NaCl as reported in the literature approach the volumes of chloroplasts swollen by HCHO and KMnO_4. Some problems concerning sampling difficulties because of wide size ranges and skewed distributions are discussed.
