Differential mRNA Transcription during the Induction of Freezing Tolerance in Spring and Winter Wheat

Gene expression at the mRNA level was studied during cold hardening and dehardening of freezing tolerant winter wheat (Triticum aestivum L. cv. Fredrick and cv. Norstar) and freezing sensitive spring wheat (T. aestivum L. cv. Glenlea). Polyadenylated RNA isolated from the crown and leaf tissues was translated in a wheat germ cell free system and the acidic and basic in vitro products were resolved in the first dimension by isoelectric focusing and nonequilibrium pH gradient electrophoresis followed in the second dimension by SDS polyacrylamide gel electrophoresis. The results showed that cold hardening conditions induced two groups of mRNAs. The first group was comprised of 23 mRNAs (coding for polypeptides with mol wt in the order of 16 to 200 kDa) which were induced and expressed at higher levels during cold hardening in the freezing tolerant cultivars compared to the sensitive one. After dehardening, their level of expression returned to that of unhardened controls indicating a positive correlation between their level of expression and the capacity of each genotype to develop freezing tolerance. This suggests that the proteins encoded by these mRNAs may play an important role during the development of freezing tolerance. The second group represents 24 mRNAs expressed at similar levels during hardening in the three genotypes. These mRNAs may code for polypeptides required for cellular adjustment during growth at low temperature and therefore are not associated with the development of freezing tolerance. These results suggest that the development of freezing tolerance is associated with specific gene expression which is regulated at the transcriptional level.
