Combined exposure of fixed tomato protoplasts to 0.2 N HCl at room temperature, heating at 70℃ in 2 × SSC for 30 min, and digestion with 10 μg/ml proteinase K allowed localization of viroid RNA by in situ hybridization, without destruction of cellular structures. The technique also allowed localization of viral RNA in tobacco protoplasts.
