The influence of the cations Ca^<2+> and Mg^<2+> on the structure of oat prolamellar bodies (PLB), set free from etioplasts and inside intact etioplasts, was examined. The results presented give evidence for a relation between PLB-structure, its transformation and the presence of divalent cations, especially Ca^<2+>. Under in vitro conditions the structure of isolated PLBs and of PLBs in intact etioplasts is stabilised by calcium. Deprivation of divalent cations by ethylenediaminetetraacetic acid (EDTA) or ethyleneglycol-bis(β-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA) led to a breakdown of PLBs to structures very similar to those found in plastids after short time illumination of etiolated leaves. Thus it seems reasonable, that Ca^<2+> is an important factor in stabilising PLBs and in the regulation of the in vivo light triggered transformation of PLBs.
