Organization and Comparison of Chlorophyll-Protein Complexes from Two Fucoxanthin-Containing Algae: Nitzschia closterium (Bacillariophyceae) and Isochrysis galbana (Prymnesiophyceae)

Three classes of discrete chlorophyll (Chl) a-containing protein complexes were isolated and characterized from both the Chl a-Chl c_1, c_2-fucoxanthin containing diatom Nitzschia closterium and prymnesiophyte Isochrysis galbana. For both algae, the highest molecular weight complexes (> 120 kDa) spectrally resembled photosystem I complexes isolated from other plant groups. Complex I was dominated by Chl a while yellow carotenoid was minor and little or no Chl c or fucoxanthin was evident in the chromophore. Second, spectrally similar light-harvesting Chl a-Chl c-protein complexes (81-82 kDa) were isolated which were highly enriched in Chl c and accounted for 48-80% of total solubilized Chl c. Third, both algae contained at least 2 distinct light-harvesting Chl-fucoxanthin protein complexes (49-98 kDa) which, with the exception of complex IV in N. closterium, contained varying amounts of Chl c. Overall results revealed general biochemical similarities between the complexes isolated from these two chromophytes (Chl c-containing algae) and those isolated previously from two dinoflagellate chromophytes which contained only Chl c_2 and had peridinin instead of fucoxanthin as their major light harvesting carotenoid [Boczar and Prezelin (1987) Plant Physiol. 83: 805-812].
