ATP concentration increased rapidly during the first 6 h of imbibition in cotyledons of cucumber and mung bean seeds. The increase was strongly inhibited by 1-h treatment of tissues with cyanide in both species. Carbonylcyanide m-chlorophenylhydrazone, an uncoupler of oxidative phosphorylation, showed little effect during the first hour of the treatment, but its in-hibitory effect on ATP synthesis became significant after 3 h. Mitochondrial fractions prepared from 6-h-old cucumber cotyledons were capable of phosphorylating ADP to ATP. These results suggest that mitochondrial oxidative phosphorylation may be involved in ATP synthesis during the early hours of imbibition in both cucumber and mung bean seeds.
