Senescence in Detached Oat Leaves II. : Identification and Changes in the Activity Levels of Aminopeptidases

Five aminopeptidases have been identified in the leaves of 7-day old oat seedlings. Three aminopeptidases were able to hydrolyse L-Met βNA, and two were active towards L-Arg βNA. Only one of the enzymes (APl) could cleave a variety of amino acid residues. The enzymes active toward L-Met βNA, L-Arg βNA and L-Ala βNA had pH optima 7.6, 8.2 and 8.3 respectively. All the enzymes were fully active in the presence of various catalytic inhibitors tested, except 1,10-phenanthroline which inhibited the activity of all the aminopeptidases, but only at a very high concentration (10 mM). The activity levels of three types of aminopeptidases tested in the crude leaf extract decreased during the senescence of oat leaves.
