Separation of Protochlorophylls Esterified with Different Alcohols from Inner Seed Coats of Three Cucurbitaceae

Reversed-phase high-performance liquid chromatography was used to separate protochlorophyllide esters isolated from inner seed coats of three Cucurbitaceae. At least 17 protochlorophylls esterified with different alcohols were separated on an octadecyl silica (ODS) column from purified pigments of pumpkin (Cucurbita moschata) and balsam pear (Momordica charantica) seeds with a single elution using 100% methanol within 22 min. The separation was done according to the numbers of carbon atoms in the esterifying alcohol of the protochlorophyll molecule with high resolution and reproducibility. Gas chromatographic analysis of the pigment-esterifying alcohols confirmed the presence of a number of protochlorophylls esterified with different alcohols. This method permits rapid, efficient and quantitative detection and identification of chlorophylls in complex mixtures.
