Membrane-bound ATPase associated with plasma membrane and soluble ATPase associated with the cytoplasm were prepared from shoots of Salicornia pacifica var. utahensis by sucrose density gradient centrifugation. The isolated ATPases were tolerant to high concentrations of NaCl. The Km for membrane-bound ATPase was 1.75 mM and for soluble ATPase, it was 1.4 mM. The relative effectiveness of divalent cations for stimulation of membrane-bound ATPase was Mg^<2+> > Fe^<2+> > Mn^<2+> > Co^<2+> > Cu^<2+>. Soluble ATPase activity was stimulated by Ba^<2+> > Ca^<2+> > Mg^<2+> and was inhibited by Cu^<2+>, Zn^<2+>, Co^<2+> and Fe^<2+>. The compounds N,N^1-dicyclohexylcarbodiimide, NaF and ADP, did inhibit the ATPases but ouabain, triphenyltin hydroxide, sodium azide, indoleacetic acid and abscisic acid did not inhibit the ATPases from S. pacifica var. utahensis.
