A subcellular fraction enriched in aleurone grains isolated in glycerol from aleurone layers of wheat endosperm specifically and reversibly bound GA_1-(^3H). Specific binding of GA_1 to other fractions including spherosomes, nuclei, mitochondria, and plasma membranes was negligible. The K_d of binding to aleurone grains was 1.5 μM and the number of specific binding sites 0.45 pmoles per mg protein. The presence of Ca^<++> ions was absolutely required for binding. Abscisic acid which inhibits giberellin action in vivo prevented specific GA_1-binding in vitro. GA_1-binding to aleurone grains is important to the primary action of the hormone which may involve mobilization of reserves from the aleurone grain-spherosome complex for utilization in membrane biogenesis.
