The in vivo data presented here are strong evidence for the involvement of citrate cleavage enzyme in lipid synthesis in developing soybean cotyledons. The incorporation of ^<14>C from citrate into crude lipid fraction in vivo had a pH optimum of 4.5; was linear with time; had a temperature optimum of 35℃; and was inhibited by (-)-hydroxycitrate. The point of citrate cleavage was between carbons 3 and 4 of the citrate molecule and therefore ^<14>C was incorporated into crude lipid fraction from citrate-5-^<14>C but not citrate-1-^<14>C or citrate-6-^<14>C.
