1. Based on changes in DNA content per whole fruit and whole fruit weight during development, the development of Japanese pear fruit (cultivars Hosui and 93-3) was divided into cell division, pre-enlargement, enlargement and ripening stages. 2. A climacteric rise in respiration with ethylene evolution was recognized although it was not very marked. 3. The ratio of pectinic acid to total pectin content increased with ripening. Total pectin content on a DNA content basis increased clearly in the pre-enlargement and ripening stages, but roughly remained constant in the cell division and enlargement stages. 4. Changes in the activities of cell wall-degrading enzymes, i.e. endocellulase, exocellulase, polygalacturonase, pectin methylesterase and β-galactosidase, were investigated during fruit growth. The activities per fresh weight of all enzymes, except exocellulase, were fairly high in the cell division and pre-enlargement stages, decreased in the enlargement stage, and increased remarkably with ripening or overripening, except for pectin methylesterase. Endocellulase was present as an acid type and neutral types. The former was more active than the latter in the cell division and pre-enlargement stages, but with ripening the reverse was found. On the other hand, the activities, on a DNA content basis, in all the enzymes were roughly constant, not decreasing during cell division, pre-enlargement and enlargement stages, but increasing extensively with ripening. That is, the lowering of these enzyme activities per g fresh weight in the enlargement stage seemed not to be due to inactivation or stimulation of enzyme de-generation. The extensive enhancements of cell wall-degrading enzyme activities with ripening or overripening seem to be closely related to the softening or pithiness of the fruit.
