Western hemlock cultures derived from cotyledon explants of 2-5 week-old seedlings were established on a chemically defined medium containing both cytokinin and auxin. Adventitious bud formation required a relatively high concentration of cytokinin with respect to that of auxin. The growth of buds was stimulated by a culture medium devoid of all plant growth regulators. Plants were successfully established in soil by rooting shoots produced from somatic cells directly in rooting medium "Mica-Peat."
