Acid proteinase activity in extracts from leaves of wheat seedlings was studied by using haemoglobin as a substrate. Optimum conditions for extraction and assay of the enzyme(s) were developed and these methods used to study the distribution of acid proteolytic activity in various tissues. The effect of leaf age and senescence on the level of activity was also investigated and the significance of these results discussed.
