A three to five-fold increase in ANS-dependent fluorescence was observed in bacterial (M. denitrificans) membranes energized by the addition of NADH or succinate. The binding kinetics between ANS and the energized bacterial membranes was studied by fluorometrical titrations and centrifugation experiments. The fluorescence was augmented by 2.7-fold in affinity towards ANS and 1.4- to 2.2-fold in relative quantum efficiency, while no change in the number of binding sites was observed upon energization. Relative quantum efficiency change was estimated from the combined results of centrifugation which separated membrane-bound ANS from free ANS, and fluorometric titration of membranes with ANS.
